Introduction: Increased use of antibiotics may potentially increase Antimicrobial Resistance (AMR) occurence. To overcome one solution for this problem is by increasing antibiotic effectiviness by combination with herbs, such as Alang-alang. Combination of crude extract of Alang-alang with Amoxicillin and Chloramphenicol has been done previously, however it has never been done with active compounds. This study was conducted to determine the effect of antibiotic inhibition when combined with semi-polar fraction (F18-23) against S. aureus and E. coli.Methods: Clear zone of single antibiotics and fractions in combination each other were measured using the plate diffusion test method and interpretation based on the Ameri-Ziaei Double Antibiotic Synergism Test (AZDAST) method. Phytochemical identification were carried out by TLC sprayed using Dragendorff reagent, formaldehyde and FeCl3, and observed for color change on the plate.Results: The results of the phytochemical test on F18-F23 found alkaloid compounds with Rf 0,8-0,86. Besides alkaloids phenol compounds were also found in F21 and F22, and steroid compounds on F18 and F19. Fraction 21-F23 had antibiotic activity against S. aureus, with ZOI 11,3±0,57mm, 7,7±6,80mm and 9,7±0,57mm which can be seen from the presence of inhibition zones. In addition to the three fractions, antibiotic activity was not found on S. aureus and E. coli. F21 had a stronger inhibition to S. aureus compared to F22 and F23, although the difference was not significant. The combination of F18-23 with antibiotics (with Amoxicillin and Chloramphenicol) against S. aureus and E. coli can not be distinguished.Conclusion: Fraction 21-F23 was found to inhibit S.aureus singularly but not E.coli. F21-F23 was presured to contain compounds Alkaloid and fenol. Interaction of F18-23 with Amoxicillin and Chloramphenicol in S. aureus dan E. coli is Not distinguishable.
